ESC has the ability to self-renewal and pluripotent. Embryonic stem cell-derived endoderm or hepatocyte is critical for the development of cellular therapies for the treatment of disease such as diabetes mellitus, liver cirrhosis. This study is divided into two parts : The first part, We used the SILAC methods to label of mouse ESC and endoderm cells, nanoLC-ESI-MS/MS to analyze the protein quantitative difference and identify 2289 proteins totally.In 2289 proteins,1354 proteins had 2 fold which 1184 protein expressions in endoderm were higher than mESC and 170 protein expressions were less than mESC.Part II, A new promising source of mesenchymal stem cells derived from the matrix of the umbilical cord (Wharton’s jelly). Wharton’s jelly mesenchymal stem cells (WJ-MSC) are able to differentiate into mesodermal, ectodermal and endodermal lineages including cell types of a variety of organs. A protein expressed in the human WJMSC was characterized by protein separation on SDS-PAGE gel coupled with nanoLC-ESI-MS/MS-based protein identification technology and bioinformatics analysis.We identified 4385 proteins.Especifically, we confirmed MSC surface markers by nanoLC-ESI-MS/MS in this study: CD44、CD105、CD166、CD73、CD13、CD90. 黃芳正 2010 學位論文 ; thesis 68 zh-TW
