Summary:碩士 === 長庚大學 === 醫學生物技術暨檢驗學系 === 98 === Autoimmune mechanism has been proving to play a role in the pathogenesis of Chronic Obstructive Pulmonary Disease (COPD). By ELISA and western blot, we investigate the prevalence and correlation of COPD of anti-RPP0、ANA、anti-RPL7 autoantibodies in patients plasma with COPD. Investigation of the correlation between pathology and the effects by COPD patient plasma against human pulmonary alveolar epithelial cells (HPAE cells). Then analysis of the relationship between the cytotoxicity and anti-CK18 autoantibody of plasma of patients with COPD, and observation of the HPAE cell morphology which against by patient plasma with COPD by immunofluorescense staining. In results, anti-RPP0 positive ratio is 0%, ANA positive ratio is 12.6%、control is 15.2%, anti-RPL7 positive ratio is 76.9%、control is 46.15%. By cytotoxicity assay, about 35.1% COPD patients with plasma cytotoxicity, but 0% in controls. Significant difference between patients and control (p<0.001). From neutralization assay、inacticvation of complement assay, there are without association with plasma cytotoxicity. From immunofluorescence staining results, plasma with cytotoxicity of COPD patients induced antibodies to enter HPAE cells. This results are suggested that it is worth for studying the mechanism of cytotoxicity of plasma of COPD patients.
