Upon binding to its ligand Delta or Jagged, Notch receptor induces signaling which regulates neural developmental events including maintenance of neural stem cell, inhibition of neuronal differentiation and promote glial determination. The zebrafish her (Hairy/E(spl)-related genes) genes are the potential target genes of the Notch signaling pathway. We had previously identified her8b genes as one of the her family in zebrafish, but the function of her8b in the neural development remains unknown. In the present study, I found that her8b is expressed in distance neural cells/tissues during development in zebrafish embryos, suggest her8b may participate in the formation of nervous system. Knock-down of her8b by antisence morpholino, resulted in eye and pericardial edema, curved trunk and abnormal inflation in swim bladder, which suggest the reason was abnormal development in early nervous system. Besides, the knock-down of her8b increase the expression of the proneuronal marker neurog1, zash1a and postmitotic neuronal marker Hu but the proglial marker GLAST was decreased, and neural stem cell marker sox2 was no inferences. However that overexpression of her8b mRNA doesn’t affect the morphology and postmitotic cells marker Hu, which suggest her8b sufficient to inhibits neurogenesis and require for gliogenesis. In addition, her8b transcripts decreased dramatically in notch deficient in viii ix mibta52b mutant line and dominant negative suppressor of hairless in injected embryos.
