The antiproliferative activity of the brown alga, Sargassum duplicatum, were tested in this study. S. duplicatum were extracted with water (WE), ethanol (EE), and supercritical carbon dioxide (SFE), respectively. The antiproliferative activity were test in human colorectal adencarcinoma cell line HT29. And, the morphology observation and cell cycle analysis were used to future elucidate the cytotoxic mechanism of S. duplicatum. SFE and EE of S. duplicatum exhibited cytotoxicity on HT29 colon cancer cell. The morphological characteristic of cell death, such as cell number decreasing and segments of cell bodies, was observed in HT29 cells treated with EE and SFE for 48 h. In addition, the HT29 cells treated with WE and EE showed progressive accumulation in G0/G1 phase, and decrease of the population in S phase at 48 h. On the other hand, SFE increased accumulation in S phase, and decrease of the population at G0/G1 phase. It appears that have different effects on cell cycle distribution induced by these extracts.
