Also, TGF-beta can promote breast carcinoma cells migration. There is already known that overexpression of hyaluronan synthase in breast carcinoma cells will synthesize high level of hyaluronan. Because high level of hyaluronan will also promote tumorigenicity and metastasis of breast carcinoma cells, we want to understand that if there is a correlation between hyaluronan and TGF-beta. We studied in three human breast carcinoma cell lines : MCF7, MDA-MB-231, Hs578T. We used real timr-PCR, western blot, HA ELISA to study TGF-beta- induced different isoform hyaluronan synthase and hyaluronan expression in transcription level and protein level. We demonstrated that TGF-beta increase HAS1, HAS2 expression and hyaluronan synthesis in MCF7 cell line and increase HAS2 expression and hyaluronan synthesis in Hs578T cell line. These results suggest that TGF-beta promoted breast carcinoma cells migration may be related to TGF-beta induced HAS1, HAS2 expression and produces high molecular weight hyaluronan in breast carcinoma cell line.
