By using host selection method, we have isolated a virus of Sulfolobus yangmingensis strain YM (SYV) and a virus of Sulfolobus tokodii strain TW (STV). By using plaque assay, we have isolated a virus and its host. Based on 16S rDNA sequence, we have identified its host to be Acidianus manzaensis strain E. We named this virus of Acidianus manzaensis strain E Acidianus manzaensis virus (AMV).SYV is an oval-shaped virus. The size of SYV virion is from 71-117 nm in length and from 47-59 nm in width. SYV virion contains three proteins and a 17-kb DNA genome. Part of the genome sequence of SYV is similar to the ORFs of Sulfolobus spindle-shaped virus 5. STV is a filamentous virus.The size of STV virion is 1333-1750 nm in length and 16 nm in width. STV harbors a double-stranded DNA genome of 18 kp and six proteins. AMV is a spindle-shaped virus. The size of AMV virion is 58-77 nm in length and 12.5 nm in width. AMV virion contains 4 proteins. AMV can form plaques on a lawn of Acidianus manzaensis strain E.
