However, the effect of CTGF on cytokine secretion in human CD14+ monocytes is mostly unknown. Here we found that CTGF increased the secretion and expression of IL-8 (Interleukin-8) in human CD14+ monocytes. Transcriptional inhibitor (Actinomycin D) inhibited the CTGF-induced secretion of IL-8 of human CD14+ monocytes. JNK MAPK kinase inhibitors (SP600125) and p38 MAPK kinase inhibitors (SB203580) both inhibited the CTGF-induced secretion of IL-8 in human CD14+ monocytes. CTGF stimulation increased the phosphorylation of JNK and p38. In addition, we used Chromatin Immunoprecipitation to observe transcription factors AP-1 and NF-κB both binds to IL-8 gene promoter.
