The inhibitory effect of oxidative stress by individual antioxidants has been well-known. Therefore, the purpose of this study was to investigate the effect of an antioxidant formula (AF) on thrombogenic risk in STZ-induced diabetic rat. Forty Wistar rats were randomly assigned into five experimental groups, including normal control (C), diabetic control (DC), diabetic rats with low dose (0.19 g/kg BW) of AF (L), diabetic rats with medium dose (0.375 g/kg BW) of AF (M), diabetic rats with high dose (0.75 g/kg BW) of AF (H). After 12 weeks of feeding period, concentration of fasting glucose and insulin, oral glucose tolerance test (OGTT), lipid profile (TG, TC), antioxidative markers (TAS、GSH、SOD、GPx、MDA), inflammatory markers (CRP、IL-6、TNF-α), coagulation factor VII (FVII), anticoagulation factor (antithrombin III, AT-III), plasminogen activator inhibitor-1 and von Willebrand factor (vWF) were measured. The results of this study showed that concentration of IL-6, vWF and activity of FVII were decreased significantly (p<0.05) in diabetic rats with low dose of AF. The serum TAS and activity of renal SOD were increased significantly (p<0.05), and concentration of MDA, IL-6, TNF-α, vWF and activity of FVII were decreased significantly (p<0.05) in diabetic rats with medium dose of AF. Serum concentration of GSH and activity of renal GPx tended to be increased in diabetic rats with medium dose of AF. The serum TAS were increased significantly, and concentration of MDA were decreased significantly (p<0.05) in diabetic rats with high dose of AF. However, GSH, IL-6, TNF-α, CRP, vWF and activity of SOD, GPx, FVII and AT-III were not different between high dose and medium dose of AF. In conclusion, antioxidative formula may have beneficial effects to ameliorate antioxidative stress, chronic inflammation, hypercoagulable state, and hence decrease the risk to develop thrombogenic complications in STZ-induced diabetic rat.
