Summary: | 碩士 === 中國文化大學 === 生物科技研究所 === 97 === Wild tomato L3683 (Solanum hirsutum Humb & Bongl), L2159 and L0994 (S. esculentum var. cerasiforme), L1947 and L6138 (S. peruvianum Mill), and tomato ASVEG#6 (S. esculentum Mill) were used as the experi- mental materials. Seedlings of 45~50 days were treated at 23℃ and 38℃with both non-flooding and flooding stress separately, for 0, 24, 48, 72, 96 hours. Plant growth and chlorophyll fluorescence were investigated. Differen- tially expressed leaf proteins of 24 and 72 hours treatments were analyzed with Proteome LabTM PF-2D and Matrix assisted laser desorption/ionization-time of flight mass spectrometer (MALDI-TOF MS).
In stressed leaves of L0994, chlorophyll fluorescence declined more seriously than other lines/cultivar. The light reaction of L0994 was the most sensitive to heat/flooding stresses. According to chlorophyll fluorescence data, stress-tolerant L3683 and ASVEG#6, stress-sensitive L0994, and in-between L6138 were selected for differentially-expressed proteins analyses. The protein in L3683 stressed leaves treated for 72 hours were respectively identified.
Comparing treatments of flooding stress at 23℃, non-flooding and flooding at 38℃ to non-flooding at 23℃, 8, 4, and 2 proteins increased; while 5, 9, and 13 proteins decreased separately in L3683. In L0994, 7, 0, and 0 proteins increased; while 4, 14, and 14 proteins reduced separately. In ASVEG#6, 1, 1, and 0 proteins increased; while 6, 6, and 6 proteins decreased separately. In L6138, 3, 11, 3 proteins increased; while 5, 2, and 7 proteins decreased separately.
