An amperometric uric acid biosensor based on gold electrode was successfully developed in this research. A three-electrode strip was fabricated using thin films technique and lift-off technique to pattern the electrodes. The Ti layer and the gold layer were deposited by sputtering on the glass substrate. After that, the reference electrode and insulation layer which were used to define electrode area were deposited using thick film technique and then gold nanoparticles were deposited on the working electrode by electroplating technique. No sooner, a thioctic acid monolayer was formed by SAMs technique and the functional group of TA monolayer was activated by EDC/NHS. Eventually, Uricase was successfully immobilized on modifying the working electrode area. The range of Uric acid concentration with a good linear relation was between 0.05 to 0.5 mM. The regression coefficient of R = 0.99991, the sensitivity of the Uric acid biosensor was 37.73 μA/cm2/mM, and the average coefficient of variation was 3.68%. The interference effect from ascorbic acid (0.17mM) was observed during the interference tests.
