MS2 without and with chloroform, glutaraldehyde or paraformaldehyde treatment were examined by a TEM after negative stained with potassium phosphotungstic acid. Chloroform and glutaraldehyde could inhibit MS2 infection on E. coli, but induced a poor quality of electron micrographs. Paraformaldehyde could inhibit the MS2 replication, but not inhibit attachment. After paraformaldehyde treatment lots of MS2 were found on the pilus of E. coli in a very good quality of micrograghs. With EM micrographs and 3D microstructure after EMAN reconstruction MS2 was proved to be an icosahedron. Without any chemical treatment MS2 showed a hole in five-fold microstructure, although with paraformaldehyde the hole was blocked. The results suggested that MS2 after paraformaldehyde treatment could attach pilus, but not transfer RNA into E. coli and replicate. The 3D microstructure of phage MS2 showed an icosahedron with an attaching mouth for transferring RNA into host.
