Summary:碩士 === 國立臺灣海洋大學 === 水產養殖學系 === 97 === This study is aimed to study mitochondrial manganese superoxide dismutase (mtMnSOD) cDNA sequence from the haemocyte of white shrimp Litopenaeus vannamei and kuruma shrimp Marsupenaeus japonicus and the tissue expressions. This study is also aimed to examine the expression of mtMnSOD when white shrimp and kuruma shrimp were injected individually with Vibrio alginolyticus and β-glucan . The mtMnSOD cDNA of L. vannamei consists of 1216 bp with 49-bp 5’ UTR, 507-bp 3’UTR, and 660-bp open reading frame (ORF) which encodes 220 amino acid. The predicted molecular mass of the translated protein is 24.1 kDa with an estimated pI of 7.95. The mtMnSOD cDNA of M. japonicus consists of 1173 bp with 40-bp 5’ UTR, 470-bp 3’UTR, and 663-bp open reading frame (ORF) which encodes 221 amino acid. The predicted molecular mass of the translated protein is 24.2 kDa with an estimated pI of 7.27. The mtMnSOD deduced amino acid of L. vannamei showed similarity of 98% to that of Fenneropenaeus chinensis. The mtMnSOD deduced amino acid of M. japonicus showed similarity of 92% to that of L. vannamei. Both L. vannamei mtMnSOD and M. japonicus mtMnSOD all expressed in haemocytes、eyestalk、intestine、muscle、gill and hepatopancreas. Both mtMnSOD transcripts of L. vannamei and M. japonicus increased after 6 h following V. alginolyticus injection indicating activation of innate immunity. Both mtMnSOD transcripts of L. vannamei and M. japonicus increased after 12 h following β-glucan injection indicating activation of innate immunity.
