In particular, large quantities of lignocelluloses in agricultural waste, which as does not compete with foodstuffs of biomass source, its development has drawn greater attention. The current commonly used methods of chemical pre-treatment, because of its high concentration of chemical substances, which has the adverse effects for wide range of levels. Therefore, ultrasound is the center methods for pre-treatment in this study. In this study, it use the common agricultural waste(Pineapple leaves, rice hull and rice straw)from Pingtung areas as raw materials. Buffer solution and Trichoderma reesei crude enzyme be used as the base solution, to solid-liquid ratio 1/10 (w/v), for 30 minutes, 25 ℃ ultrasound of pre-treatment, then analysis the percentage of reducing sugar from it, and watch the structural change by SEM. To different concentrations (3 %,5 %) to 50 ℃, 24 hours of enzyme hydrolysis, analysis the conversion rate of reducing sugar and enzyme activity. The results showed that different base solution at pre-treatment under ultrasound, Trichoderma reesei crude enzyme has higher reducing sugar of release than buffer solution (rice hull:> 0.1 mg / ml; rice straw:> 0.6 mg / ml; pineapple leaves: > 0.4 mg / ml). From SEM observation of samples after pre-treatment structure, compared to no pre-treatment, in the Trichoderma reesei crude enzymes for pre-treatment, the fragmentation phenomenon more than the former. In enzyme hydrolysis, rice husk at the 5 % (no pre-treatment) has the highest conversion rate 10.84 %, because its structure closed, so no high values for other way of pre-treatment. Rice straw at the 5 % (Trichoderma reesei crude enzymes for pre-treatment) has the highest conversion rate 56.8 %.
