According literatures, Cordyceps possesses various biological and pharmacological activities. The polysaccharides of Cordyceps have been studied and found to be capable of interaction with the receptors of immunocytes, induction of a series of signal transduction, and then resulting immunocytes activation and replication. Besides, scientists have indicated that the biological functions of polysaccharides with various structures may vary. In this study, ethanol precipitated polysaccharides of Coydyceps myrmecophila were prepared. The crude polysaccharides were applied to an FPLC equipped with a size exclusion column to fractionate the polysaccharides of C. myrmecophila in different molecular weights. The molecular weight, saccharide compositions and structure linkage of each fractionated polysaccharide were determined. The cellular proliferation, phagocytic activity and cytokine expression of immunocytes boosted by various fractionated polysaccharide were also analyzed. The results indicated that the molecular weight range of the fractionated polysaccharides was between 100 and 170 kDa. The compositions of the polysaccharides were mainly glucose with α-(1→4) and α-(1→6) linkages, the rest was glucose with α-(1→3). According to the results of immuno activity analyses, the polysaccharides of C. myrmecophila may induce immunocyte and T cell and B cell multiplication proliferation up to 1.4 to 1.7 folds, comparing to that of the control set without adding any polysaccharides. In addition, the polysaccharides of C. myrmecophila induce phagocytic activity of macrophage up to 13.9 to 21.8%, comparing to that of the control set as well. The polysaccharides of C. myrmecophila may induce cytokine (IL-2、IL-1β、TNF-α) express to enhance 230 to 560 pg/mL、40 to 95 pg/mL、80 to 126 pg/mL, comparing to that of the control set as well respectively, besides, the polysaccharides of C. myrmecophila in different molecular weights express IFN-γ to enhance 139 to 182 pg/mL with Con A, comparing to that of the control set as well. In conclusion, the fractionated polysaccharides of C. myrmecophila had a similar saccharide composition and structure feature, however, the fractionated polysaccharides of C. myrmecophila to possess immunomodulation effects degree.
