(Vitaveae), a perennial woody vine plant, is widely distributed in Taiwan, and has been used in traditional folk medicine. Air-dried stems of A. brevipedunculata var. hancei were extracted with methanol. The MeOH extract was suspended in H2O, and then partitioned sequentially using EtOAc and n-BuOH. In preliminary antioxidant, cytotoxic and anti-inflammatory assays, the EtOAc layer of the stems of A. brevipedunculata var. hancei exhibited cytotoxic and antioxidant activities, that led us to investigate the bioactive principles from this plant. The EtOAc layer was further purified by column chromatography, HPLC, and recrystallization to yield forty-three compounds, including fourteen triterpenoids, nine aromatics, eight steroids, three flavonoids, three stilbenes, three oligostilbenes and three lignans. Among these, 6α-hydroxy-5α-stigmastan-3-one (Ste8), 5,7-dihydroxy-2-[2-(4-hydroxyphenyl)ethyl]chromone (F3) and (+)-9'-O-trans-coumaroyl-5,5'-dimethoxylariciresinol (L3) were isolated from the natural source for the first time. Their structures were determined by spectral analysis and chemical conversion. Betulinic acid (T2), resveratrol (Sti1), 3,4',5-trihydroxybibenzyl (Sti2), p-hydroxybenzaldehyde (Ar1), and gallic acid (Ar9) showed significant cytotoxic activities against SK-Hep1 cell line with IC50 values of 16.3, 92.1, 114.8, 340.1, and 140.6 μM, respectively. Furthermore, resveratrol (Sti1), syringic acid (Ar7), protocatechuic acid (Ar8), gallic acid (Ar9), and syringaresinol (L1) showed significant DPPH free radical scavenging activities with EC50 values of 65.1, 66.8, 46.1, 13.2, and 45.4μM, respectively.
