Our results showed the Pregna exhibited more growth inhibitory effect on HL-60 than other tested human leukemia cells, such as U937, THP-1, Jurkat. Pregna inhibited cell proliferation and induced apoptosis in a concentration- and time-dependent in HL-60 cells. Treatment of HL-60 cells with pregna, it resulted in dissipation of mitochondrial membrane potential, release of mitochondrial cytochrome C into cytosolic, activation of caspases 9 and -3, disruption of DNA integrity, and induction of apoptosis Moreover, treatment with Pregna caused a rapid loss of intracellular glutathione content and stimulation of reactive oxygen species (ROS) generation. N-acetylcysteine (NAC), an antioxidant, and the pan-caspase inhibitor, z-Val-Ala-Asp-fluoromethyl ketone (z-VAD-FMK), significantly inhibited Pregna-induced cell death. Taken together, it suggests that induction of apoptosis by Pregna may be modulated by generation of ROS and activation of caspase activity.
