The main advantage of high-voltage pulse electroporation is that it does not require any medium, and this method can transfer the DNA sequence or drug into the cell. The cell membrane is stimulated by current pulses in the electroporation, so reversible small holes in the cell membrane are created, and the material in the buffer was moved into the cells by diffusion. The purpose of this study was to combine the bio-medical and optical technology, and the chip was fabricated by MEMS process for transfer gene, and combined with a programmable power supply control, and it was integrated by the programmable power supply with LabVIEW, so that power supply generated the electrical field. One needs to complete the transfection effectively, and enhance the gene transfection by adjusting the voltage, pulse number and time. The 293T cells were tested in the experiments. From the electroporation result, it showed that the best buffer is Hypoosmolar (Eppendorf), the best electrical field is 1600 V/cm. The best pulse number and time are two times of high electrical field 1600 V/cm with 0.6 ms, and 3 times of high electrical field 800 V/cm with 1.2 ms, respectively.
