Quercetin was reported to have significant anti-inflammatory activity because of its direct inhibition of several initial processes of inflammation. This study is to examine whether quercetin inhalation could decrease the inflammatory response in adult Sprague-Dawley (SD) rats with sepsis -induced lung injury. Sepsis-induced lung injury was induced by cecal ligation puncture (CLP) in adult SD rats. Quercetin inhalation was given in different time interval after CLP. Neutrophil,pro-inflammatory cytokines (IL-1β、IL-6、TNFα) from serum and bronchoalveolar lavage fluids (BALF) were analyzed. Lung tissue was stained with hematoxylin and eosin to survey the severity of lung injury. We found the neutrophils in blood serum increased significantly at 9th hour and 18th hour in SD rats after CLP; however, delayed enhancement of neutrophils in BALF was observed at 12-24th hours. After quercetin inhalation (20 mg, 40 mg, 60 mg ) during 1st-3rd hour and 7-9th hour respectively, pro-inflammatory cytokines (IL-1β、IL-6、TNFα) in BALF was decreased significantly in study groups (p<0.05). Pathological examination, lung injury was also found to be less severe comparing with those in control groups. However, neutrophil and pro-inflammatory cytokines in peripheral blood did not decrease significantly after quercetin inhalation. We concluded that quercetin inhalation is effective to reduce the inflammatory response and subsequent tissue injury in a sepsis-induced lung injury model.
