Due to the different clinical symptoms after infected by classical swine fever virus, CSF is still not fully eradiated and sporadic cases remain. The major prevention method of CSF in Taiwan is CSF vaccine injection. The two types of CSF vaccines currently used in Taiwan are lyophilized lapinized Philippines coronel (LPC) vaccine and lyophilized lapinized CSF tissue culture (LPC-TS) vaccine. LPC and LPC-TS vaccines both are attenuated vaccines, so the amount of live virus in the vaccines has a great impact in generating the immune potency. Therefore, the purpose of this study is using real-time quantitative polymerase chain reaction (PCR) to develope a high sensitivity, high specificity, and rapid method for CSF vaccine detection. In this study, we have successfully established a NS5B gene TaqMan real-time PCR system that can detect 1.8x105-3x101 CSFV. We hope this detection system can be used for CSF vaccine production monitoring. Further application is used to monitor the efficiency of vaccination and as a reference for the reinforcement injection which will increase the breeding rate of pig farms and to reduce production costs and to increase the income of pig farmers.
