Summary: | 碩士 === 中原大學 === 生物科技研究所 === 97 === Emodin (1,3,8-Tri-hydroxy-6-methyl-anthra-quinone) is a tyrosine kinase p65lck inhibitor which isolated from Rhei Rhizoma. Previous studies have documented many effects of emodin, including anti-tumor, anti-inflammatory, cell cycle inhibition, apoptosis, and reactive oxygen species (ROS) generation. Recent researches show that emdoin also affects on neovascularization and fibrosis, but there is no study showed the effects on blood cells. In this investigation, we used zebrafish model to determine whether there are any injury effects by emodin. Zebrafish embryos that were treated with different concentrations of emodin have found morphological differences. We investigated embryos which treated with over 2μM have morphological changes. Although, emodin suppress angiogenesis, but in our study there have no effects on vascular tube formation were found in zebrafish model. We found that emodin had hazardous effects on zebrafish’s blood cell. There is less or no blood was found in emodin-treated embryos for two days, so we thought maybe emodin affects on mesoderm. And then, we used whole-mount in situ hybridization to determine whether emodin has specific effects on blood cells that development from mesoderm. So far we investigated emodin would not affect expression of gata1, ndrg1, mpo and pu.1. Emodin may not suppress synthesis of blood cells but probably affects on blood cell viability.
