Using Serratia marcescens swarming as a research model, we found a two component system RssA-RssB that is involved in repression of swarming motility. Recently, we observed that RssA-RssB signaling regulates the initiation of swarming in S. marcescens CH-1. Nevertheless, the underlying mechanism remains unclear. This study finds that RssA-RssB represses iron acquisition gene pvc, which is required for the production of pseudoverdine. Therefore iron availability may signal RssA-RssB system to modulate swarming motility. This study shows that the influence of swarming motility in S. marcescens CH-1 by extracellular ferric iron depending on RssA-RssB two-component system. Environmental iron concentration alters RssA-RssB two-component system signaling state. Furthermore, RssA-RssB coordinates pseudoverdine production and the timing of initiation of swarming. The pseudoverdine-deficient mutant S. marcescens ΔptxR-pvc influenced the swarming behaviour of S. marcescens through modulation of duration of RssA-RssB signaling. This study proposes that iron availability acts as a signal to modulate the RssA-RssB signal transduction systems to influence the swarming behavior of S. marcescens. H. C.
