In this study, we use raman spectroscopy to analyze the normal cervical intraepithelial squamous cell(with 125 cells) and cervical precancer which include low grade squamous intraepithelial lesion(LSIL) with 44 cells and high grade squamous intraepithelial lesion(HSIL) with 16 cells. We detect the nuclear of the single squamous cell on unstained and non-closing thinprep pap smear. According to the spectral analysis, the raman peak intensities at 1240cm-1(CN stretch, NH bending amideⅢ band)、1578 cm-1 (C=C stretch, nucleic acids)、1605 cm-1 (CO stretching)、1660-1665 cm-1 (C=O stretch, amideⅠ) increase with the grade of cervical precancer.
All spectra were normalized to the intensity of the 1004 cm-1 peak and grouped by pathology. For further analysis, we apply the area ratio 1240/1004、1600group/1004 cm-1 of all spectra to form the two-dimensional scatterplots. The linear discriminator separates three group and also carry out cross validation classified rate 90%.
