Summary:碩士 === 國立陽明大學 === 解剖暨細胞生物學研究所 === 96 === Enolase is a ubiquitous, glycolytic enzyme (2-phospho-Dglycerate hydrolase) that catalyzes the dehydration of 2-phospho-D-glycerate (PGA) to phosphoenolpyruvate (PEP) in glycolysis. In normal cells, ENO1 is a cytosol-localized glycolytic enzyme. Recent studies demonstrate that ENO1 has multiple cellular functions in addition to glycolysis. Immunohistochemistry revealed significant overexpression of ENO1 in the tumor sections as compared to extremely low level of this protein in the adjacent normal tissues. In addition, expression of ENO1 and patient’s survival rate was found to be correlated. ENO1 was also found in MCF-7, MDA-MB361 and SK-BR3 by western blotting analysis.To investigate the role of ENO1 in the breast cancer cell , over-expression of ENO1 in MCF-7 and down-regulation of ENO1 in SK-BR3 were established. MTT viability assay , migration and invasion analysis revealed that over- expression of ENO1 promoted cell proliferation, migration and invasion of MCF-7 cells. Down-regulation of ENO1 decreased the ability of the cell proliferation, migration and invasion in SK-BR3 by MTT viability assay , migration and invasion analysis. In conclusion, ENO1 plays an important role in breast tumor cell metastasis.
