Summary:碩士 === 大同大學 === 生物工程學系(所) === 96 === Haloferax mediterranei is used in this study as an extremely halophilic archaebacterium for red pigment (hmERP) production. In this study, we investigate the composition, characteristic structure and molecular weight of components in hmERP. The optical properties of hmERP were analyzed by UV/VIS Spectrophotometer, and shown that the absorption wavelength of hmERP are 527, 494, 468, 388 and 371 nm with maximum wavelength at 494 nm. We also found that the absorption wavelength of hmERP decayed regularly under light irradiation. From the infrared absorption spectrum, the molecular structure of hmERP contain hydroxy (-OH) and C=C double bond. After separating by reversed-phase High Performance Liquid Chromatography (HPLC) at C18 column with 70% acetone and 30% water as the mobile phase, the hmERP contains at least six compounds. The molecular weight 741.6(M+H)+ of peak A (major peak with shortest retention time) in hmERP was obtained by LC-APCI-MS. Based on the IR spectrum, molecular structure and molecular weigh analysis, we conclude that component in peak A of hmERP is same as Bacterioruberin (a C50 carotenoid derivative).
