Our lab has previously found that NP can induce a rise of cytosolic calcium concentration ([Ca2+]c),and inhibited the [Ca2+]c rise under the stimulation of acetylcholine receptors (AChR). In this study, we investigate the effects of serum albumin on the changes of [Ca2+]c in bovine adrenal chromaffin cells and human neuroblastoma SH-SY5Y cells to clarify the physiological possibility of NP’s interference characters. We found 1% serum albumin almost inhibited the [Ca2+]c rise induced by NP 5μM, and counteracted the inhibition of NP on nicotinic AChR, while 0.025% serum albumin had partial effects. Similar effects of albumin occurred on NP-induced the changes of mitochondria calcium concentration ([Ca2+]mt) and NP-induced cell death. In RT-PCR measurements, NP decreased the gene expression of TEF2 and increased the gene expression of HSPA8 in the presence of serum, while decreased in the absence of serum. In the last part of my study, we utilize the character of NP interfering [Ca2+]c to detect the NP residual concentration after bacterial degradation. We found the toxicity was increased. Then, 2-butyl-1-octanol, the biodegradation product of NP, had characters to interfere [Ca2+]c and might the possible candidate to remain the cytotoxicity of NP.
