The result showed that the G.lucidum may produce manganese peroxidase and laccase under two cultivation ways, and have efficient degradation ability for nonylphenol.Under the liquid state culture with 2 mg/L nonylphenol, low nitrogen source (0.22 g/L) to compare with the high nitrogen source (2.2 g/L) can enhance 4 times of nonylphenol degradation half-life. The ablity of degradation nonylphenol (20 mg/L) was clearly increased by adding tartaric acid (20 mM), MnSO4 (0.5 mM) and citric acid (20 mM), during 2 days of culture, degradation of 20 mg/L nonylphenol can reach 82%, 77% and 34%, but 1% Tween 80 was inhibited degradation. During solid state culture, the laccase activities in the different matrixs is wheat atraw (10232 U/g), sawdust (128 U/g), rice bran (0.19 U/g) and manganese peroxidase is wheat atraw (736 U/g), sawdust (9 U/g), rice bran (0.1 U/g) respectively. The optimum conditions for produced enzyme was mixed the sawdust and wheat atraw at ration of 1:4 and 5 ml/g content water. Polyacrylamide gel electrophoresis of crude enzyme by concentrated extracellular culture fluid confirms that the different culture conditions and supplements can induce different molecular weight laccase isoenzymes.The liquid state cultures showed two bands ( 50 kDa ) with laccase activity, and the solid state cultures produces three laccase isoenzymes, ranging from 40 and 60 kDa. Two raise ways may gather the high molecular weight polysaccharide( > 1000 kDa ), but polysaccharide production from liquid state were highest ( 526 mg/g). The enztme supernatants from solid state culture was completely degraded 20 mg/L nonylphenol、bisphenol A and phthalate esters, present of mixture pollutants ( 20 mg/L PAEs、BPA ) still had the good degradation ability for nonylphenol. Remazol brilliant blue R ( 0.01 mM ) is the best redox mediator, may promotes 20 mg/L nonylphenol degradation reach 1.9 and 13.3 times within eight days inculation. The sum up, G. lucidum strain CCRC 36021 under the solid state culture can produce an enzyme to degrade nonylphenol. In future, we can use the solid state matrix waste as the biological filter bed in the waste water, after the processing, to become organic compost has the value and saves many wastes processing cost, achieves goal of the G.lucidum production zero pollution.
