Summary: | 碩士 === 靜宜大學 === 食品營養研究所 === 96 === Lactoferricin (Lfcin), an antibacterial peptide, is released from N-terminal of lactoferrin by pepsin digestion. Lactoferricin II (Lfcin II) , exhibited on the outer surface of the amino-terminal lobe of lactoferricin, includes an 11 amino acids of amphipathic region. In this study, two recombinant lactoferricin, Lfcin II and Lfcin II-(His)6, were expressed from lactoferricin cDNA integrated Pichia pastoris expression system, created and donated kindly by Master Huang, Guo-Ting. The recombinant Lfcin II and Lfcin II-(His)6 were separated purified using CM-Sephadex cation exchange chromatography & HisTrapTM affinity chromatography. The Lfcin II and Lfcin II-(His) 6 were revealed ACE- inhibition activity with IC50 values of 121.91 μg/ml and 226.24 μg/ml, respectively. The inhibition mechanism of both Lfcin II and Lfcin II-(His)6 were non-competitive with Ki values of 0.12 mg/ml and 0.22 mg/ml, respectively.
