碩士 === 國立東華大學 === 海洋生物科技研究所 === 96 === The benthic Prorocentrum strain PL040104002 were isolated from the coral reef area of southern Taiwan, and were further studied for their chemical nature by batch-culture technique. The extracts were separated by using bioassay-guided separation strategy. Seven polyketides 1~7 were isolated from the dichloromethane layer, and two marcolide 8, 9 were isolated from the butanol layer. OA-D10b (1) and OA-D10a (2) are two okadaic acid diol esters. Prorocentin-4 (3) is a new class of polyketide with two tetrahydrofuran rings. Formosalide A (4) & B (5) are the 17-membered macrolide with two tetrahydrofuran rings, two diene moieties, and two branched methyl and five hydroxyl groups. (E) - 2,7-dimethyleneoct-4-ene-1,8 diol (6)、(Z)- 2,7-dimethyleneoct-4-ene-1,8 diol (7) are two diol moieties of 1 and 2, respectively. The structures of these compounds were elucidated on the basis of NMR and mass data. In the biological MTT cytotoxicity assay test, compounds 1, 2, 4, 5 exhibited potent cytotoxicity on the cell line of CCRF_CEM . The LD50 values of the compounds were between 0.04~1.56
