Viruses have been isolated from the gastrointestinal and respiratory tracts of chickens, especially in viral arthritis and pale bird syndrome in poultry. In our previous study, 1-week-old chickens were inoculated by foot pad with 2408 strain, the mortality caused by 2408 is closed to 100%, but that by S1133 strain is different. Therefore we studied relationship between the cytokines and clinical disease in vitro after inoculation of virous virus strain. In our study, chicken macrophages were prepared from the blood of specific-pathogen-free (SPF) Leghorn chickens. Macrophages were treated with S1133, 2408 and 919 strains at 5x105TCID50 for 0, 8, 16 and 24 hours. Total RNA was isolated from macrophages. IL-1, IL-6, IL-8, IL-12, IFN-α, iNOS and K60 were detected by RT-PCR at various periods of times after infection. The results were expressed as the ratio of mean count of cytokines expression level to that of β-actin expression level. Statistical analysis was performed using ANOVA and Tukey test by SPSS. Results showed that the mRNA expression of all of cytokines had a significant increase in 8 hours after the infection in all of strains. After 8 hours of infection, mRNA expression of IL-1, IL-6, IL-12, IFN-α and iNOS were increased in S1133 strain but IFN-α was decreased in 2408 strain. IL-1, IL-6, IL-8, IL-12, IFN-α, iNOS and K60 were decreased after 8 hours of infection and K60, IL-12 were increased after 16 hours of infection in 919 strain. IL-1, IL-8 and K60 were increased after the infection of S1133 strains, but only IL-1 and K60 had a significant increase (p<0.05). After 8 hours of infection, IL-12 was increased (p<0.05) in S1133 strain. The results were related to lymphocytes and neutrophils were infiltrated in synovial membranes. IL-6, IL-8 and K60 had a significant increase (p<0.05) after the infection of 2408 strains. Acute phase protein secreted by liver induced by IL-6 and IL-8. Hepatitis and necrosis resion in liver were correlated with IL-6 and IL-8.
