The first task is the development of a suitable quantitative method for following the attachment yield. After methods developments by Ultraviolet/Visible Spectrophotometer (UV/Vis.) and High Performance Liquid Chromatography (HPLC), an optimized quantitative method was obtained. It is a RPLC method with wavelength of 272 nm, mobile phase of Methanol 35%: DI-water 65%. This method is proved to be the fastest, effective and least interfering way for following the complex extracted from the natural herb. The kinetics of the attachment was followed up by HPLC quantitative method. Three different carbon nanotubes were used. First one is carbon nanotube in 10-40 nm porosity with 95% purity. Second one is carbon nanotube in 40-60 nm porosity with 95% purity. Third one is carbon nanotube in 40-60 nm porosity purified by microwave digestion. The natural herb chosen for this study is Polygonum chinense L. which helps skin regeneration and demonstrated Staphylococcus aureus, Shigella, Typhoid bacillus, Colon bacillus pharmacology function. But Taiwanese folk soak their wound skin in the heated vinegar solution, which is not a scientific or sanitized way. The difference between the two kinds of 40-60 nm carbon nanotubes, before and after the purification by microwave digestion, can be observed obviously by Scanning Electron Microscopy. The HPLC system was used for proving the fact that both more polar and less polar portions, existing in small amount in the complex, were able to be attached by nanotube. The kinetics conclude that 10-40 nm porosity with 95% purity and 40-60 nm porosity with 95% purity , 40-60 nm porosity purified carbon nanotubes can attach the less polar portion up to 35.95% ,38.99% and 30.08% of natural extract respectively at room temperature. The carbon nanotubes can attach the polar portion up to 45.45%, 51.81% and 45.45%. Each gram of carbon nanotubes can attach up to 32.48 g, 35.39 g and 27.33 g of Polygonum chinense L..
