Dried tuber of T. giganteum, known as Bai fu zi, has been commonly used as traditional Chinese medicine for the treatment of headache, hemicrania and other syndromes. A standardized protocol for the establishment of callus culture using the root explants and mass- propagation of the plant is reported here. The optimum condition for callus induction is using a medium of full strength MS salts, 1.0 mg/l 2,4-D, 3% sucrose and 0.9% agar, and then growing in dark at 25±1°C for 8 weeks. And as for the maintaining of callus, medium containing full strength of MS salts, 1.0 mg/l 2,4-D and 0.2 mg/l BA were used. For the mass-propagation of the plant, the medium should contain full strength MS salts supplemented with 3% sucrose, 0.9% agar and 40 g/l banana pulp. To study the function of hemagglutinating activity of Bai fu zi, a number of cultured explants were collected. Crude extracts obtained after phosphate buffer saline (PBS) and 40-60% ammonium sulfate extraction followed by dialysis were used for the treatment of erythrocytes. The result showed a remarkable increasing in the hemagglutinating activity and the light even enhance such activity. The protocol established in this thesis can provide a new and promising source of T. giganteum for further biochemical study and medical applications.
