The extract prepared from the roots of Rhei Ehizoma, Coptidus Rhizoma and Scutellariae Radix (SHXXT) was demonstrated more excellent hepato-protective activity than the single herb whish used individually as indicated in this studies. The SHXXT are sorted three groups: water-boil, micro-powders and nano-powders. Liver fibrosis induced in male ICR mouse by oral administration with 5% chloroform twice a week for 8 weeks. At the same time, SHXXT (58 and 116 mg/BW) was administered intragastrically. The results was investigated that low dosage of Nano-SHXXT alleviated liver fibrosis induced by chloroform in mouse. Additionally, there was a remarkable improvement of CAT, SOD, GSH and GSH Px (GPx) activities. The results from in vitro studies showed MMP-2, TIMP-1, Bcl-2 mRNA expressions decreased. However, the increase Bax, Fas mRNA expression increased in hepatic stellate cells (HSCs) activated by TGFβ1. These results suggested that Nano-SHXXT could not only inhibited significantly the progression of hepatic fibrosis induced by chloroform, but inhibited the proliferation of activated hepatic stellate cells by TGF-β1.
