Because new viruses generated by reassortment may be extremely virulent, understanding the reassortment of viral genes is essential. Bioinformatic formulas for analyzing amino acid sequences of H1N1 and H3N2 virus subtypes reveal that NS1 protein signatures are more distinct than those of other internal genes. To identify possible restrictions on NS gene reassortment, this study applied a reverse genetics method to generate WSN recombinant viruses that NS gene is replaced by NS genes of different virus subtypes. The WSN recombinant viruses containing NS genes of A/TW/141/02 (H1N1) grew faster than those containing NS genes of A/TW/3446/02 (H3N2). A CAT-ELISA assay was also performed to measure the effect of NS proteins from different subtypes on viral polymerase activity. The A/TW/3446/02 NS protein exhibited the most dramatic enhancement of polymerase activity. The signature results of this difference are still under investigation.
