Studies have demonstrated that betel quid chewing is associated with increased risk of oral cancer. Areca nut is the major substance of areca quid, and arecoline is the major ingredient of areca nut. Areca nut extract (ANE) and arecoline have been shown to induce DNA strand breaks and the main cause of cellular transformation and carcinogenesis. However, there was still not clear about whether apoptosis would be caused by areca nut. Since immediate early response gene X-1 ( IEX-1 ) can be induced by a lot of stimulus and furthermore regulate the cell proliferation and cell apoptosis, the specific aim of this study is to investigate the role IEX-1 in apoptosis induced by ANE and arecoline. The results have shown that the cell viability of oral cancer cell lines decreased after the increase of time and dose of ANE and arecoline treatment. The elevation of IEX-1 mRNA was also in accordance with the increase of apoptosis after ANE and arecoline treatment. Furthermore, IEX-1 protein was also found in the cytoplasm of OECM-1cell line after ANE treatment. The activated caspase-9 and caspase-3, cleaved PARP were also found to be induced by ANE. In conclusion, cell apoptosis induced by ANE and arecoline may be associated with IEX-1 expression.
