Summary: | 碩士 === 國立陽明大學 === 熱帶醫學研究所 === 95 === This thesis reports two immunodominant antigens, the 53kDa excretory/secretory glycoprotein (gp53) and tropomyosin (TsTM) from Trichinella spiralis in their molecular characteristics and immunodiagnosis. Characterization of gp53 indicated it might be a metabolic enzyme for nucleotides and had a special signal peptide which could cause toxic effect on host cell.Data also showed that TsTM had a α-helic structure.The great similarity with tropomyosin of other species cause cross reaction with other helminths. TsTM showed highly antigenic after immunized with mice.On the other hand, purified gp53R(recombinant gp53) were able to disintegrate plasmid DNA in the presence of Mg+2 and Ca+2, the same ability as native excretory/secretory antigens, indicates it as a DNase. The evaluation of the immunodiagnostic test using Trichinella-infected sera、naïve sera (pigs and mice) and schistosoma-infected sera (mice) showed while using gp53R as antigen, no false positive occurs and the antigen could detect trichinellosis as early as 2nd weeks post infection (p.i.). TsTMR(recombinant TsTM) could detect 2nd week p.i. in pig and 1st week p.i. in mice. However, using TsTMR as antigen, cross reaction between Schistosoma and Trichinella infected mice sera was observed in the immunodiagnostic tests. Detection rate at 100% were achieved by these two proteins after 3rd weeks p.i. in pigs, these results indicated that these two antigens could satisfy the need of high-specificity and sensitivity serodiagnosis in swine trchinellosis.
