Thirty male Sprague-Dawley rats were divided into five groups: control (C), exhaustive exercise (E), exhaustive exercise with low dose (0.1875 g/kg BW) (EL), middle dose (0.9375 g/kg BW) (EM) and high dose (1.875 g/kg BW) GT supplementation (EH) groups. After 30 days, the rats of exercise groups underwent an acute exhaustive running test on a motorized treadmill. After the running test, all rats were sacrificed immediately. The blood, muscle and liver samples were collected. The energy metabolism, the muscle glycogen content and blood glucose were significantly decreased and blood urea nitrogen (BUN) was increased in E group (p<0.05), compared with C group. Compared with E group, the free fatty acids (FFA) concentration in EL group, blood glucose concentration in EM group, and the liver glycogen content in EM and EH group were significantly decreased (p<0.05). On the other hand, the running time was obviously prolonged in EL and EH group than that of E group (p<0.05), and it significantly in EH group than that of EL group (p<0.05). For the antioxidative status, the activities of superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GRD) and GSH/GSSG ratio in the erythrocytes were obviously reduced (p<0.05). There were no significant differences in SOD, CAT, glutathione peroxidase, GRD activities, and GSH/GSSG ratio in EL, EM and EH group, when compared with E group. TBARS level of EH group was apparently decreased than that of E group (p<0.05). About the tissue damage, the activities of aspartate transaminase (AST), alanine transaminase (ALT), creatine kinase (CK) and lactate dehydrogenase (LDH) in the plasma were significantly raised in E group (p<0.05), when compared with C group (p<0.05). The apoptosis of muscle cell in E group was more apparent than in C group. The plasma AST, ALT, CK and LDH activities in EL, EM, and EH group had no changes, when compared with E group. The apoptosis of muscle cell in EL, EM, and EH group was less apparent than in E group. The results show that low dosage supplementation of Ganoderma tsugae before exhaustive exercise may maintain glucose homeostasis, muscle glycogen content and attenuate apoptosis of muscle cell to prolong running time. 楊素卿 2007 學位論文 ; thesis 105 zh-TW |
