Taiwanese has the habit to drink Chinese alcoholic medicine during the daily life to maintain and/or improve health. However, the toxicity induced by drinking of Chinese alcoholic medicine has been occurred. Therefore, the safety for the long term administration of Chinese medical alcohols is valuable to be evaluated.
Method:
It’s going to investigate the safety for long term administration and Longevity Tonic of Kinmen alcoholic medicine which was produced by ourselves following the principals of Chinese medical alcohols from Committee on Chinese Medicine and Pharmacy (Department of Health, Executive Yuan, Taiwan). The analysis of ourselves-made Chinese medicinal wine will be enforced by High Performance Liquid Chromatography with two standards of the most component Chinese medicine of Kinmen alcoholic medicine. According to scientific literatures, there’s a lot of antioxidative-related efficacy of the component Chinese medicine of Kinmen alcoholic medicine being investigated.
Therefore, We first evaluated the in-vitro antioxidaive ability by the experiment of DPPH (α, α-Diphenyl-β-picrylhydrazyl ) scavenge activity. Afterward we evaluating the acute and chronic toxicity individually by feeding Kinmen alcoholic medicine to rats or mice ,and the long-term effects of herbs composed of these medicinal wines will be investigated additionally. The 90-days feeding toxicity study and the pathological diagnosis was to evaluate the toxicity might be caused by the long-term administration of Kinmen alcoholic medicine to the experimental animals.
Furthermore, we additionally investigate the activity of antioxidative enzyme of the rat’s liver.
Results & Discussion:
There are no obvious acute toxicity in Wistar rats received oral administration of self-made Kinmen alcoholic medicine at 15 ml/kg/day for 14 days. However, not only the eyes and tails of Wistar rats were redness but also frequent pruritus and the light color faces were observed in rats received oral administration of commercial or privately Kinmen alcoholic medicine for 90 days. The plasma levels of glutamic-oxaloacetic transaminase and glutamic-pyruvic transaminase were increased in rats received a long-term administration of home-made Kinmen alcoholic medicine at 1.67ml/kg/day; The plasma levels in triacylglycerols is reduced when taking low dose of Kinmen alcoholic medicine for 30 days ( short-term).However, the plasma levels in cholesterol as well as the ratio are in normal range. In addition, the levels of blood urea nitrogen and creatinnine were in the normal range in rats received a long-term administration of home-made Kinmen alcoholic medicine, indicating glomerulus were not damaged. The pathological photograph of liver shows that at least occurred steatosis or even worse situation in the group of Kinmen Yi-Shou Medicinal Wine or 25% alcohol rats. In the evaluation of the activity of antioxidative enzyme of the rat’s livers, the experimental results showed that Kinmen alcoholic medicine could improve the release of superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase activity in tissue/organs and it’s benefit for scavenging free-radicals.
As the above, if we can modify the concentration of alcohol for producting a Kinmen alcoholic medicine which can reduce the damage resulting by alcohol and active the antioxidative enzyme of animals. The modified Kinmen alcoholic medicine will be benefit for prolong life and improve the health.
