Many papers reported that Lingzhi possesses anti-tumor, anti-microbial, anti-viral, anti-allergy, anti-hypertension and anti-aging. The most important biological activities of Lingzhi are polysaccharides and triterpenoids. The aim of this research is to study the effect of cell growth, polysaccharide concentration, high molecular weight of polysaccharides and triterpenoid productions by various conditions (strains, carbon sources and additives) to establish the optimal conditions for G. lucidum. The results showed that it could not enhance the cell growth, polysaccharide concentration and high molecular weight of polysaccharides when G. lucidum CCRC36111 and A. blazei CCRC36814 presented simultaneously. However, polysaccharide concentration was enhanced from 24 mg/g to 38 mg/g. The high molecular weight of polysaccharides was increased when G. lucidum CCRC36111 and A. blazei CCRC36814 presented simultaneously. Comparison with different carbon Sources, the cell growth of G. lucidum CCRC36111 was increased from 9.20 g/L to 16.74 g/L by the addition of maltose. However, Glucose as carbon source enhanced the polysaccharide concentration of G. lucidum CCRC36111 from 25 mg/g to 104 mg/g, and high molecular weight of polysaccharides and triterpenoid also increased. For the addition of different particle sizes of A. blazei powder, we found that the larger particle size of A. blazei, the higher cell growth and polysaccharide production. However, high molecular weight of polysaccharides was increased when the A. blazei particle size was lower than 0.5 mm. For the addition of different additives, we found that the best number of cell growth by addition of 1.0 g A. blazei powder. Polysaccharide concentration increased from 24 mg/g to 78 mg/g by addition of 1.0 g sawdust. The high molecular weight of polysaccharides had the best production by the addition of 1.0 g L. edodes powder. The result also exhibited addition of various additives could induce G. lucidum CCRC36111 to product different triterpenoid. The results showed that the maximal production for high molecular weight of polysaccharides was at day 20, and decreased at day 30. We observed the structure of polysaccharides by NMR, and indicated that the major activities of polysaccharides (C1 - C3 and C1- C6) decreased and disappeared from day 20 to day 30.
