This bacterium was identified as Alcaligenes sp. strain Yen (AY744384). It has a plasmid with 18,400 bp and had been succesfully transformed into competent E. coli HB 101 cells. Positive colonies were visualized forming pits on the modified C-10 agar plate. The enzyme was purified from culture supernatant by ammonium sulfate precipitation and gel filtration column chromatography method. The optimal activity for purified enzyme was at pH 7.5 and at temperature 45 ℃. The enzyme had a specific agarase activity of 3,200 U/mg. Estimation of the agarase molecular mass by SDS-PAGE was 101 kDa. The agarolytic bacterium have the highest agarase activity among all previous reported studies.
