Description
Summary:碩士 === 國立臺灣海洋大學 === 生物科技研究所 === 95 === Systemic Lupus Erythematosus is a chronic-immune disease affected many somatic organs such as joints, skin, kidney and et al by auto-antibodies, including anti-DNA and anti-nuclear protein antibodies. Auto-antibodies, 9B6 and 9D7 recognizes ribosomal P protein and DNA, respertively. To identify one cross-reactive antigens of these antibodies, mass spectrometry was performed after immunoprecipitation and SDS-PAGE. 28 candidates of antigens were analysed by LC-MS. Glucose-regulated protein 78 (GRP78), one of candidate, were characterized by co-immunoprecipitation and western blotting using 9B6 antibody. Our results indicated that this strategy may be used to identify the complex recognized by auto-antibodies and to study the pathogenesis of SLE.
