Graduate date: 2007/07/12 Degree Conferred: Master Name of student: Chein-Wei Chen Adviser: Hso-Chi Chaung The contents of abstract in this thesis: Pattern recognition receptors (PRR) include two superfamilies, Toll-like receptors (TLR) and C-type lectin receptors. The expressions of PRRs on immune cells function to recognize different pathogen associated molecular patterns (PAMP) and thus induce different signal pathways in cell, which in turn causes the secretion of cytokines to regulate the acquired immune responses. Thirteen different TLRs have been demonstrated in human, however, there are only a few literatures on porcine TLRs. In this study, the effects of poly I:C on TLRs expressions of three porcine antigen-presenting cells including alveolar macrophage (AM), immature bone marrow haematopoietic-derived dendritic cells (BMHC-imDC), mature DC (BMHC-mDC) were investigated. The results showed that the TLR 3 and 8 gene expressions in AM were higher than those in imDC and mDC. No significantly higher expressions of TLRs were observed in mDC. Furthermore, the stimulation of poly I:C may inhibit the gene expressions of TLR 7 and 8 in AM and TLR8 in imDC but enhance the expression of TLR3 in AM.
