For the harmful effects from the agglutinin activity, we tried to use the non-cytotoxic dosage with heating treatment to study the immunomodulatory effects on the human moncytes and the inhibitory effect on U 937 cell lines. It has shown that both the CEL and heating treated CEL (HCEL) exerted no cytotoxic effect on U937 at 30μg/mL. However, The MNC-CM( conditioned medium from monocyte cell) stimulated by both the CEL and HCEL at 20 μg/mL after 24 hours treatment can inhibit the growth of U937 and expression dose-dependent, respectively. The differentiate effect of CEL-MNC-CM can also observed as morphological change, from blast to matured or macrophages to about 7.5 ± 2.5 % in 20 μg/mL;NBT reduction up to 24.4 ± 3.2 % and expression dose-dependent. It is interesting that there is not any signification difference between HCEL and control group. The TNF-α、 IL-1β and IFN-γ can be found in CEL-MNC-CM, but only TNF-α and IL-1β in HCEL-MNC-CM. The obvious increment of human monocytes can also be observed after the stimulation by CEL and HCEL. We have confirmed that the proliferation of monocytes come from the immunomodulatory effects of CEL and HCEL on monocytes. Some kinds of cytokines or lymphokines were induced by monocytes to activate lymphocytes, substantially; the activated lymphocytes interact with monocytes to induce the proliferation of monocytes.
