Melanocyte transplantation is a method to solve this problem. However, skin color varies from locations of the body and person to person. Furthermore, the distribution of melanocytes and the amount of melanin production were thought to be the crucial factors in skin pigmentation. Thus it is critical to determine the amount of melanocytes to be transplanted for treatment of hypopigmentation. Objective: In this work, we try to investigate the parameters crucial for skin pigmentation in terms of “L” value determined by chromameter, melanocyte numbers, and melanin production, and find the relationships among them. Methods: The human melanocytes and keratinocytes were cultured and purified from skin sample. All of these parameters mentioned above were collected and analyzed based on different human foreskin samples in vitro. The ratio of melanocytes to epidermal cells is determined by using flow cytometry. The melanin production on the basis of aliquot numbers of melanocyte was measured by using spectrophotometer. Results: We found a significant correlation (R = 0.848) between L value and the amount of aliquot melanin production multiplied by the ratio of melanocytes to epidermal cells.
