The Human Umbilical Vein Endothelial Cell Line (ECV304)) was cultured on the biomedical material, polydimethylsiloxane PDMS. Some parameters such as the alternates of the working solution, the styles of PDMS templates, regulative frequency and collagen smearing and so on were changed in this study. The object of this research is to find an adaptive measurement model for adhesion force of the cell. According to the experimental results, the microstructure on PDMS substrate can help to improve the uneven problem of the collagen smearing by PDMS hydrophobic property. The cell cultured on the region of microstructure can reduce the collage expend and confirm that the measured cell is on region of the smearing collagen or not. Three different cultured areas on the PDMS substrate, such as plane surface, large area, and small area, were conducted to do the measurement of endothelial cell adhesion force with dielectrophoresis force. The results found that the cell cultured on the large area of substrate which has 2 mm microstructure interval had better adhesion force than that on the small area of substrate which has 100 �慆 microstructure interval. And the cell cultured on the large area can obtain nice growth and faster expand, so that the cultured cell on the large area substrate is suitable to measure cell adhesion force. The 2% glucose solution which own lower electricity is suitable to be used for the measurement of adhesion forces.
