The purpose of study is investigated to measure the cell membrane
stiffness for the red blood cells of anemia and normal rats using AFM .
There were three categories such as the cell membrane stiffness
measurement at the different solution concentrations for the red blood
cells of normal rats, the blood cell membrane stiffness difference between
the anemia and normal rats and the relationship between the membrane
stiffness and physiology conditions for the red blood cells of rats.
At differences osmosis concentration, the average membrane stiffness
of red blood cells for normal rats on the 2.2%NaCl solution is
165.72×10-3N/m, the average membrane stiffness of red blood cells for
normal rats on the 0.9%NaCl solution is 44.07×10-3N/m and the average
membrane stiffness of red blood cells for normal rats on the 0.45%NaCl
solution 17.94×10-3N/m. This result shown the solution concentration
effected the membrane stiffness measurement. From the experimental
results, the average membrane stiffness (45.3×10-3N/m) for the red blood
cells of normal rats is less than that of anemia rats (109.2×10-3N/m). It
means the blood cells of anemia rat represents brittle and it is not easy to
passing through a blood capillary. The relationship between the
membrane stiffness for the blood cells and physiology conditions of rats
such as RBC count, HGB and HCT, are shown negative relationship and
the determinant coefficient (R2 ) are greater than 0.8. It means the
relationships between the membrane stiffness and physiology conditions
of rats are obvious negative.
