This method using solid-solid extraction (SPE) and N,O-bis (trimethylsily) trifluoroacetamide with 1% trimethylchlorosilane derivatization, followed by GC-MS analysis using dextromethorphan as the internal standard. The method was linear from 62.5~2000 ng/mL. The limit of quantification and limit of detection were 62.5 and 31.25 ng/mL, respectively. The intraday precisions range from 6 to 10 %。The interday preciosions range from 4~11 %。This method was further subjected to an interlaboratory study. Nine laboratories participated in a study to analyze 3 samples in duplicate. Estimates for the repeatability and reproducibility of the method, expressed as relative standard deviations of the results of the determination of pentazocine in urine, were 3.0~5.2 % and 6.8~10.3 %, respectively. The HORRAT values among those 3 samples in duplicate are range from1.1 to 1.5. Therefore method reproducibility as normally would be expected according to the guidelines of AOAC.
