Description
Summary:碩士 === 中興大學 === 獸醫微生物學研究所 === 95 === Avian reovirues are important pathogens of poultry industry, because they can cause considerable losses of the farming economy. There are very few viruses have the ability to cause cell-cell fusion, and ARV is one of them. ARV usually induces arthritis, malabsorption syndrome and chronic respiratory disease. The feces and organs of chickens were collected from Taichung, Nantou, Changhua and Yunlin. The samples described above were inoculated into CEF (chicken embryo fibroblast) to isolate the virus, and detected by RT-PCR method. Sequencing results were compared with the data saved in Genebank to confirm the ARVs existing. In the same time, we used a pair of primers specific to ARV S1 gene to amplify the target sequences, and followed by genotype analysis. This study successful isolated five field ARVs, temporally assigned numbers from 1 to 5. The S1 gene of the five field strains have the closest relation with GEL13b98M (isolated from Germany), NC/SEP-R61/03 and 138 (isolated from US). The five field strains isolated from this study, ARV vaccine strains and the ARV strains isolated from Taiwan in 1970~1992 could be classified into different lineages.
