Description
Summary:碩士 === 中興大學 === 獸醫微生物學研究所 === 95 === Waterfowl circoviruses are emerging diseases of waterfowls and these viruses could be divided into goose circovirus (GoCV) and duck circoviruses (DuCV). The infection of waterfowl circoviruses might lead to immuno suppression and increase the incidence of other diseases. This laboratory previously used the E. coli expression system to express the replication protein (Rep) and capsid protein (Cap) of GoCV and DuCV but found that the only the Rep protein but not the Cap protein could be expressed. The goal of this study is to use the baculovirus expression system to express the Rep and Cap proteins of GoCV and DuCV and to use these proteins as antigens to detect the antibodies against GoCV and DuCV in geese and ducks. The result shows that Rep and Cap proteins of GoCV and DuCV could all be successfully expressed in baculovirus expression system. Moreover, Western blot assays using these proteins as the antigens showed that the antibody titers against GoCV and DuCV were low in sera collected from geese and ducks in the field. This result suggests that the infection of GoCV and DuCV might produce only a low level of antibody response. This is the first report on the expression of the full-length Rep and Cap proteins of GoCV and DuCV.
