Description
Summary:碩士 === 中興大學 === 材料工程學系所 === 95 === Using biopolymer as the chelating agent, we have synthesized CDHA(calcium deficient hydroxyapatite) by wet-chemical method. Through SEM (scanning electron microscopy) observations, the morphology of CDHA microspheres are hydrangea-like porous. From the specific surface area/porosimetry & chemisorption analysis, the porous particle exhibit the specific surface area of 24.3 m2/g, and the micropore and mesopore volume of 0.23 cm3/g. Taking advantage of its high drug loading ability, the tests absorption and release for gentamicin were carried out. Because of the resorption of HA, a slow and constant release of drug over a prolonged period was found. The in vitro cytotoxicity assay indicated that porous CDHA microspheres can improve the proliferation and differentiation of osteoblast-like cell, because of the helpful peptite and the bioactivity and osteoconductivity of HA. The high gentamicin-loaded and injectable CDHA microspheres have been successfully prepared to achieve drug sustaining release. This achievement will be very helpful to resolve the tough problems of chronic osteomyelitis and reveal the advantages of micro-invasion and osteointegration.
