The surface micropattern can align cells and the microporous wall can allow the nutrients and wastes to be transported effectively. In this study, the symmetric/asymmetric poly(D,L-lactide)(PLA) substrates with/without surface microgrooves were fabricated by phase transition process. Furthermore, the symmetric/asymmetric PLA substrates covered with a top layer of chitosan with/without surface microgrooves were developed. The behaviors of glioma C6 cells and neural stem cells (NSCs) on these substrates were examined at 24 and 72 h. It was found that C6 cells and NSCs had highest percentage of alignment on the chitosan/PLA composites , lower percentage of alignment on symmetric substrate with microgrooves and lowest percentage on asymmetric substrate with microgrooves. Comparing C6 cells and NSCs, the degree of NSCs alignment was higher than that of C6 in all groups. Additionally, the degree of cell alignment increased with time in all groups. The neurotrophic factor gene expressions on these substrates were studied. The neurotrophic factor gene expression was up-regulated on the chitosan/PLA composites with mirogrooved surface, compared to the microporous PLA substrates at 24 h. However, the gene expression of neurotrophic factors was higher on PLA than on chitosan/PLA composites at 72 h.
